    : ELISA (enzyme-linked immunosorbent assay) is a blood test that detects antibodies based on a reaction that leads to a detectable colour change in the test tube. It was the first practical approach using enzyme tracing of the reaction. It is a sensitive technique used to detect small quantities of specific proteins by antigenic means. The HIV-ELISA is commonly used as a initial screening test. Originally, the antigen for these tests were prepared from infected T-lymphocyte cultures; however it was impossible to exclude all lymphocyte-derived material from these preparations, and therefore false-positive reactions occurred. Now-a-days Western or immunoblotting has been used extensively as a confirmatory assay.
