    Explain insertional inactivation. Name 2 insertional inactivation techniques.

    Answer:

    Selectable marker are antibiotic resistance gene and gene coding for enzyme P galactosidase. When foreign DNA is inserted into these marker genes, they inactivate and lose their activity. This insertional inactivation leads to screening of transformed cells from non-transformed ones.
    (i) Insertional inactivation of antibiotic reaction gene allowing transformed cells to grow only on antibiotic for media.
    (ii) Inactivation of gene coding for enzyme \[\beta \] galactosidase which gives blue colour in presence of chromogenic substrates in culture media.
