                    Isolation of restriction endonucleases by Nathans and Smith (1970) made it possible to cut DNA at specific sites. Restriction enzyme can cut both strands of DNA when foreign nucleotides arc introduced in the cell. They cleave DNA to generate a nick with a 5' phosphoryl and 3' hydroxyl terminus.
