                     Molischs test is not used for testing of proteins. It is used in testing of carbohydrate. Molischs test 1 mL aqueous, solution of organic compound, +2 drops of 10% ale. solution of a-naphthol + shake well + 1 mL of cone. \[{{H}_{2}}S{{O}_{4}}\to \] a reddish violet ring at the junction of two layers is formed which indicates the presence of carbohydrate. Miltons test Millons reagent is prepared by dissolving mercurous and mercuric nitrates in nitric acid. On adding Millons reagent to a solution of protein, a white ppt. is obtained which turns brick red on boiling. Biuret test Protein is dissolved in strong caustic soda solution. A few drops of dil. \[CuS{{O}_{4}}\] solution are added to above solution, then a bluish violet colour is obtained. This test is due to the presence of ?NH?CO? group in protein. Ninhydrin test Proteins, having free ?COOH and ?\[N{{H}_{2}}\] groups, give an intense blue colour with ninhydrin. Note - Molischs test is given only by those proteins which a carbohydrate.
