  • question_answer
A student wishes to study the cell structure under a light microscope having 10X eyepiece and 45X objective. He should illuminate the object by which one of the following colors of light so as to get the best possible resolution?

    A)  Yellow

    B)         Green

    C)  Blue

    D)         Red

    Correct Answer: C

    Solution :

Resolving power or resolution is the ability of the lens to distinguish fine details and structure. Specifically it refers to the ability of the lenses to distinguish between two points at a specified distance apart. Resolving power depends on two factors: (i) Wavelength of light used for illumination (ii) Power of objective lenses \[\text{Resolving power }=\frac{\text{Wavelength of light}}{\text{2}\,\text{x}\,\text{NA}}\] Since the limit of resolving power of a microscope is fixed by the structure of light, the shortest wavelength of visible light will give the maximum resolution. Among yellow, green, red and blue light colour. Blue (500 nm) have shortest wavelength so, it will give best resolution.
